Melanotan II (MT-II; Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH₂) is a synthetic cyclic analog of alpha-melanocyte-stimulating hormone (α-MSH). It was developed at the University of Arizona in the early 1990s by Victor Hruby and colleagues as part of a program to identify potent, metabolically stable melanocortin receptor agonists.

MT-II binds with high affinity to melanocortin receptors MC1R, MC3R, MC4R, and MC5R. Published preclinical studies have examined MT-II’s effects on penile erection via MC4R activation in the CNS (Dornan et al., 1996; Wessells et al., 1998), skin pigmentation via MC1R, and appetite suppression via MC3R/MC4R pathways.
