Kisspeptin-10 is a synthetic decapeptide corresponding to the C-terminal ten amino acids of kisspeptin-54, the primary secreted product of the KISS1 gene in humans. The KISS1 gene was first characterized as a metastasis suppressor in 1996 at Pennsylvania State University, and its G protein-coupled receptor — GPR54, now designated KISS1R — was identified as the endogenous receptor in 2001 by independent groups including Kotani et al. and Ohtaki et al.

The bulk of published research has focused on kisspeptin-10’s role as a high-affinity KISS1R agonist within the hypothalamic–pituitary–gonadal (HPG) axis, where it acts on GnRH neurons in the arcuate nucleus to modulate pulse generator frequency and downstream luteinizing hormone secretion. Research groups at the University of Edinburgh (Millar, Roseweir, Anderson) and the Medical Research Council’s Centre for Reproductive Health have contributed substantially to this body of literature. All findings referenced here derive from preclinical models and have not been validated in controlled human clinical trials.
